We performed an arrayed comparative genomic hybridization using a cDNA spotted array, which allows the simultaneous analysis of the gene expression levels of over 5000 human genes in a single experiment. We obtained a set of genes that was differentially expressed in response to cadmium in placental tissue. Among these, we identified several genes whose expression is known to be involved in cadmium toxicity. In addition, we identified several uncharacterized genes whose expression is uniquely regulated in response to cadmium. These new cadmium responsive genes were identified as potential therapeutic targets to alleviate the adverse effects of cadmium exposure. We also identified a number of genes that are specifically regulated in response to cadmium in human placenta, but not in most other tissues. These genes would be interesting candidates for developmental toxicity studies. In order to evaluate these genes further, we have established cell lines from sheep placenta and in vitro cultured trophoblasts, which are the precursors of the placental trophoblasts. We hope to develop an in vitro model to study the effects of cadmium on the function and viability of trophoblasts. Studies of the effects of cadmium and other toxicants on gene expression in cells in vitro may be important for establishing test methods for chemicals that are difficult to study in vivo. We have also used microarrays to study the effects of cadmium on whole genome expression in human cell lines. These studies have allowed us to identify the up regulation of several genes that may be involved in the toxicity of cadmium. Although cadmium is known to affect heme synthesis in fetal cells, the mechanisms that lead to heme deficiency in cadmium-exposed embryos are not well understood. We are using microarray analysis to investigate the effects of cadmium exposure on the regulation of genes of heme biosynthesis in the developing human placenta. We have identified genes that are specifically expressed in vivo in the trophoblasts as well as genes that are specifically regulated in vitro in the cytotrophoblast and syncytiotrophoblast.
