Everything You Need to Know About Mutation Breeding

Mutation breeding is a powerful technique for creating new and improved varieties of crops. It involves exposing plant material to agents that can cause changes in their DNA, such as radiation or chemicals. These changes, called mutations, can result in new traits that are beneficial for the plant, such as higher yield, better quality, or resistance to diseases. Mutation breeding can also overcome some of the limitations of other breeding methods, such as sexual incompatibility or hybrid sterility.

In this article, you will learn about the theory and practical applications of mutation breeding, and how it has contributed to crop improvement for more than 70 years. You will also find out how to download a free PDF of Mutation Breeding: Theory and Practical Applications, a comprehensive book by A. M. van Harten that covers all aspects of this technique.

What is Mutation Breeding?

Mutation breeding is based on the fact that all living organisms have a certain rate of spontaneous mutations, which are random changes in their DNA. These mutations can be beneficial, neutral, or harmful, depending on how they affect the organism’s function and fitness. Most mutations are either neutral or harmful, and are eliminated by natural selection. However, some mutations can confer an advantage to the organism, and are passed on to the next generation.

Mutation breeding aims to increase the frequency and diversity of beneficial mutations in plants by exposing them to mutagens, which are agents that can cause DNA damage. Mutagens can be physical, such as radiation (e.g., X-rays, gamma rays, or neutrons), or chemical, such as alkylating agents (e.g., ethyl methanesulfonate or EMS), base analogues (e.g., 5-bromouracil or 5-BU), or intercalating agents (e.g., acridine orange or AO). Mutagens can induce different types of mutations, such as point mutations (changes in single nucleotides), insertions or deletions (addition or removal of nucleotides), inversions (reversal of a segment of DNA), translocations (exchange of segments between chromosomes), or polyploidy (increase in the number of chromosome sets).

Mutagenesis can be applied to seeds, pollen, buds, cuttings, or tissue cultures of plants. The treated plant material is then grown and screened for desirable traits. The plants that show improved characteristics are selected and tested for stability and performance. The selected mutants are then multiplied and released as new cultivars.

What are the Advantages and Limitations of Mutation Breeding?

Mutation breeding has several advantages over other breeding methods:

  • It can create novel traits that are not available in the existing gene pool of the plant species.
  • It can overcome the barriers of sexual incompatibility and hybrid sterility that limit conventional cross-breeding.
  • It can be applied to any plant species, regardless of their reproductive system or ploidy level.
  • It can be combined with other techniques, such as marker-assisted selection or genetic engineering, to enhance the efficiency and precision of breeding.
  • It is relatively simple, fast, and inexpensive compared to other methods.

However, mutation breeding also has some limitations:

  • It is largely dependent on chance and requires large populations and intensive screening to identify useful mutants.
  • It can induce undesirable mutations that may reduce the fitness or quality of the plants.
  • It can cause chimerism (a mixture of mutant and normal cells) in vegetatively propagated crops, which may affect the stability and uniformity of the mutants.
  • It may raise ethical and environmental concerns about the safety and regulation of mutagen-treated plants.

What are some Examples of Mutation Breeding?

Mutation breeding has been used for more than 70 years to improve various crops around the world. According to the Food and Agriculture Organization (FAO) and the International Atomic Energy Agency (IAEA), more than 3200 mutant cultivars have been officially released in over 200 plant species. Some examples of mutation breeding are:

  • The development of semi-dwarf wheat varieties

  • The improvement of barley quality by inducing mutations in genes that affect malting characteristics. These mutants have higher diastatic power, lower protein content, and better germination than normal barley varieties.
  • The creation of seedless grapefruit by inducing polyploidy in buds using colchicine. These mutants have larger fruits with fewer seeds than normal grapefruit varieties.
  • The enhancement of disease resistance in rice by inducing mutations in genes that confer resistance to bacterial blight, blast, tungro virus, or brown planthopper. These mutants have increased tolerance or immunity to these diseases than susceptible rice varieties.
  • The modification of flower color in chrysanthemum by inducing mutations in genes that affect anthocyanin biosynthesis. These mutants have novel colors such as white, yellow, pink, purple, or bronze than normal chrysanthemum varieties.
